Inventor(s): Moshal; Jeffrey M. (Allentown, PA), Libman; Michael (Allentown, PA)
Assignee: GENUS LIFESCIENCES INC. (Allentown, PA)
Application Number:16/915,488
Patent Claims: 1. A method of providing induction of local anesthesia of mucous membranes in an adult patient comprising: placing four pledgets in about 4 mL of a pharmaceutical composition comprising about 36 mg/mL to about 44 mg/mL of cocaine hydrochloride, 1.2 mg/mL to 1.4 mg/mL citric acid, 0.7 mg/mL to 1.3 mg/mL sodium benzoate, water, and more than zero and less than about 10% benzoyl ecgonine, wherein each pledget of the four pledgets has an absorbency sufficient to absorb about 1 mL of the pharmaceutical composition; placing two pledgets of the four pledgets in each nasal cavity of the adult patient adjacent to a septum of the adult patient for up to about 10 minutes to about 20 minutes, provided that the method results in an absorptivity factor for cocaine in the adult patient ranging from about 0.25 h.sup.-1 to about 1.0 h.sup.-1 of cocaine in the adult patient, provided that the method results in a T.sub.max ranging from about 0.01 hours to about 2.00 hours of cocaine in the adult patient, provided that the method results in an AUC.sub.inf in a range from about 182 ng*h/mL to about 3446 ng*h/mL of benzoyl ecgonine in the adult patient, provided that the method results in a T.sub.max in a range from about 1.00 hours to about 8.00 hours for benzoyl ecgonine in the adult patient; and removing each pledget placed in a nasal cavity of the adult patient from the respective nasal cavity, wherein each pledget removed from a nasal cavity of the adult patient retains from between about 70% to about 90% of the pharmaceutical composition; and performing a diagnostic or surgical procedure on or through the nasal cavities of the adult patient, provided that the method results in a reduction in pain equivalent to achieving a visual numeric rating score of 0 upon von Frey filament testing.

2. The method of claim 1, wherein a length of each pledget of the four pledgets is about 3.0 cm.

3. The method of claim 1, wherein the pharmaceutical composition contains not more than about 10% total impurities.

4. The method of claim 1, provided that the method results in a T.sub.max in a range of about 1.00 hours to about 4.00 hours for ecgonine methyl ester in the adult patient.

5. The method of claim 1, wherein the procedure is a nasal laryngoscopy.

6. The method of claim 1, wherein the procedure is a nasopharyngeal laryngoscopy.

7. The method of claim 1, wherein the procedure is a nasal endoscopy.

8. The method of claim 1, provided that the method results in a T.sub.max ranging from about 0.15 hours to about 1.00 hours of cocaine in the adult patient.

9. The method of claim 1, provided that the method results in an AUC.sub.inf in a range from about 1000 ng*h/mL to about 3446 ng*h/mL of benzoyl ecgonine in the adult patient.

10. The method of claim 1, provided that the method results in a T.sub.max in a range from about 1.00 hours to about 4.00 hours for benzoyl ecgonine in the adult patient.

11. The method of claim 1, wherein the pharmaceutical composition contains not more than 6.5% benzoyl ecgonine.

12. The method of claim 1, wherein the pharmaceutical composition contains not more than about 500 ppm methanol per 4 mL.
